Effets anti-inflammatoire, toxicologique et analyse phytochimique des extraits de Erica arborea

| dc.description | Erica arborea connue sous le nom vernaculaire Akhlendj, est un arbuste de la famille des Ericaceae dont les parties aériennes sont utilisées en médecine traditionnelle algérienne comme agents anti-inflammatoire, diurétique, antimicrobien et antiulcéreux. A notre connaissance, aucune étude ne combine entre l’effet anti-inflammatoire, antioxydant, toxicologique, et la caractérisation phytochimique des extraits aqueux (AQ), éthanolique (EtOH) et hydro-éthanolique (EtOH 70%) de cette plante. A cet effet, notre thématique de recherche a pour objectifs d’évaluer l’activité pharmacologique citée précédemment, l’effet antioxydant, d’établir le profil toxicologique, et de prendre connaissance des composés phytochimiques de ces extraits. L’évaluation de l’effet anti-inflammatoire in-vivo des trois extraits a été réalisée en utilisant deux modèles expérimentaux : l’œdème plantaire provoqué par la carragénine et l’œdème de l’oreille induit par l’huile de croton. L’administration orale des trois extraits (250 et 500 mg/kg) réduit significativement l’œdème plantaire et auriculaire. Cependant, l’extrait AQ montre un effet plus important. . L’activité antioxydante des différents extraits d’E. arborea a été détérminée par l’association de plusieurs tests tels que le test du DPPH, ABTS, test du blanchiment du ß-carotène et le pouvoir réducteur. Les résultats ont montré que les extraits possèdent une forte activité antioxydante qui peut être attribuée à la qualité et quantité des composés phénoliques et flavonoïdes. L’évaluation toxicologique des différents extraits a permis de démontrer que leur administration aiguë n’induit ni mortalité ni effets indésirables indiquant que la DL50 est supérieure à 5000 mg/kg. L'administration répétée (28 jours) de l’extrait AQ (500 et 1000 mg/kg) a montré une réduction significative des niveaux de triglycérides chez les rats mâles et femelles alors qu'aucune altération significative des autres paramètres biochimiques et hématologiques n'a été observée. Des dommages histo-pathologiques ont été observés dans les tissus hépatiques et rénaux des rats des deux sexes traités avec la dose 500 et 1000 mg/kg de l’extrait AQ. L'analyse HPLC-DAD-MS a permis d'identifier des acides phénolique dont l’acide chlorogénique, l’acide acide p-coumarique héxoside et l’acide 5-O-p-coumaroylquinique et cinq flavonols glycosides : myricétine pentoside, quercétine-3-O-glucoside, myricétine-3-O-rhamnoside, quercétine-3-O-pentoside et quercétine-3-O-rhamnoside. Nos extraits peuvent être considérée comme non toxiques en administration aiguë tandis que l’extrait AQ présente une toxicité modérée en administration subaiguë. Les résultats obtenus lors de l'étude anti-inflammatoire corroborent l'utilisation traditionnelle de l'extrait AQ d'E. arborea pour traiter les troubles inflammatoires. Ainsi, cette plante peut être considérée comme une source prometteuse de composés naturels qui peuvent être incorporés dans notre alimentation et les applications pharmaceutiques. | |
| dc.description.abstract | Erica arborea known as Akhlenj is a shrub, belonging to Ericaceae family that its aerial parts are used in Algerian traditional medicine for anti-inflammatory, diuretic and antiulcer purposes. To our knowledge, no studies reveal the combination between in-vivo anti-inflammatory activity, antioxidant effect, toxicological evaluation, and phytochemical characterization of the aqueous (AQ), ethanolic EtOH and hydro-ethanolic (EtOH 70%) extracts of this plant. For this purpose, this study was designed to evaluate the pharmacological activity cited above, the antioxidant effect, highlight on the toxicity of these extracts, and deepen the knowledge on and phytochemical composition of these extracts. The anti-inflammatory activity of the extracts was assessed using carrageenan induced paw edema and croton oil induced ear edema. This pharmacological investigation revealed that the E. arborea extracts possess a strong anti-inflammatory effect where the AQ extract displayed the best effect. The antioxidant activity of the different extracts have been evaluated by association of four tests: DPPH scavenging test, radical cation ABTS+, β-carotene bleaching test and reducing power. The results showed that the extracts have a strong antioxidant activity that can be attributed to the quality and quantity of the phenolic and flavonoid compounds. Acute administration of the AQ, EtOH and EtOH 70% extracts did not reveal any mortality or side effects indicating that the lethal dose 50 (LD 50) is above 5000 mg/kg. Repeated administration (28 days) of the AQ extract (500 and 1000 mg/kg) showed a significant reduction in triglyceride levels in male and female rats, while no significant changes in other biochemical and hematological parameters were observed. Histo-pathological damages were observed in liver and kidney tissues of animals of both sexes treated with 500 and 1000 mg/kg dose of the AQ extract. HPLC-DAD-MS analysis led to the identification of phenolic acids and derivatives such as chlorogenic acid 5-O-p-coumaroylquinic acid and p-coumaric hexoside acid and five flavonol glycosides: myricetin pentoside, quercetin-3-O-glucoside, myricetin-3-O-rhamnoside, quercetin-3-O-pentoside, and quercetin-3-O-rhamnoside. The extracts can be considered non-toxic in acute administration while the AQ extract shows moderate toxicity in sub-acute administration. The results obtained in the anti-inflammatory study corroborate the traditional use of E. arborea aqueous extract to treat inflammatory disorders. | dc.description.sponsorship | Erica arborea المعروفة محليا بالخلنج ، عبارة عن شجيرة تنتمي إلى عائلة Ericaceae .تستخدم أجزائها الهوائية في الطب التقليدي الجزائري لأغراض مضادة للالتهابات، مدرة للبول، مضادة للميكروبات ومضادة للقرحة. إلى حد علمنا، لم تجرى أي دراسات النشاطية المضادة للالتهاب، المضادات للأكسدة، التقدير السمي على الحيوانات ، و التحليل الكيميائي للنبات، للمستخلص المائي (AQ)، الإيثانولي (EtOH) والمستخلص الهيدروإيثانولي (EtOH 70%) لهذه النبتة. لهذا الغرض ، تم تصميم هذه الدراسة لتقييم التأثير الفرماكولوجي (الصيدلاني(٬ تقييم الخصائص المضادات للأكسدة ، تسليط الضوء على سمية هذه المستخلصات، وتعميق المعرفة في التركيب الكيميائي لهذه المستخلصات. من خلال النتائج، أبدى التقييم الفرماكولوجي لمستخلصات E. arborea تأثير معتبرا كمضاد للالتهابات، حيث أظهر مستخلص AQ أفضل تأثير مضاد للالتهابات مقارنة للمستخلصات الاخرى .تم تقييم النشاط المضاد للأكسدة لمستخلصات النبتة من خلال أربعة اختبارات: اختبار إزاحة جذرDPPH ،ABTS ، اختبار التبييض β-caroten و اختبار القدرة الإرجاعية . أظهرت النتائج أن المستخلصات المختبرة لها نشاط قوي مضاد للأكسدة و قد يعود ذلك إلي نوعية وكمية المركبات الفينولية والفلافونويد. لم تِؤدي السمية الحادة لمستخلصات AQ و EtOH و EtOH 70% الى حدوث وفيات أو آثار جانبية لدى الجرذان و هذا يشير إلى أن الجرعة المميتة 50 ( (DL 50تفوق 5000 مغ/كغ. أظهرت السمية الشبه حادة (28 يومًا) للمستخلص AQ / 500) 1000 مغ/ كغ (انخفاضًا معنويا في مستويات ثلاثي الجليسريدات لدى الجرذان الذكور والإناث، بينما لم يلاحظ أي تغيرات في المؤشرات البيوكيماوية والدموية الأخرى. كما أظهرت النتائج النسيجية المرضية بعض التغيرات في أنسجة الكبد والكلى لدى الجرذان المعاملة من كلا الجنسين بالجرعتين 500 و 1000 مغ/ كغ من مستخلص AQ . كشف تحليل التركيب الكيميائي للمستخلصات عن وجود كمية عالية من المركبات الفينولية، وأدى تحليل HPLC-DAD-MS إلى تحديد كل من حمض الكلوروجينيك -p-O-5 كوماريل كنيك و –pكوماريك هيكسوسيد وكذالك خمسة جليكوزيدات فلافونول: ميريسيتين بنتوسيد ، كيرسيتين -3-O- جلوكوزيد ، ميريسيتين- 3-O- رامنوزيد ، كيرسيتين -3 -O - بنتوسيد ، وكيرسيتين -3- O- رامنوزيد.. كخاتمة، يمكن اعتبار المستخلصات غير سامة في حالة الاستخدام الحاد بينما يُظهر مستخلص AQ سمية معتدلة في السمية شبه الحادة. تؤكد النتائج التي تم الحصول عليها مخبريا فعالية الاستخدام التقليدي للمستخلص المائي للخلنج في علاج الاضطرابات الالتهابية. يمكن أن تكون نبتة الخلنج مصدرًا واعدًا للمركبات الطبيعية التي يمكن دمجها في نظامنا الغذائي والتطبيقات الصيدلانية | |
