Isolement de bactéries extrêmophiles d’intérêt agricole productrices de molécules antifongiques

| dc.description.abstract | Salinity is one of the major factors responsible for the deterioration of soils, making them unsuitable for agriculture. The application of halotolerant and halophilic plant growth- promoting bacteria PGPR: Plant Growth Promoting Rhizobacteria) with biological control activities as crop inoculants offers a biological alternative to the use of , particularly when they are subjected to saline stress. To this end, 70 bacterial strains were from saline soils (sebkha) in arid and semi-arid zones of eastern Algeria. Three isolates were selected based on their production of bioactive molecules that promote plant growth. 100% of isolates have the capacity to fix nitrogen and produce ammonia (NH3). 53.33 % have the ability to solubilise inorganic phosphate, and 33.33% produce (HCN) and 6.67% produce IAA. Two of these isolates belong to the genus Serratia and the third is a halophilic Halomonas bacterium. These bacteria were identified based on the 16S rDNA sequence. Antagonism tests against phytopathogenic fungi were carried out. The antifungal molecules by these bacteria were identified using high- liquid chromatography. These bacteria are capable of inhibiting mycelial growth against fungi, with rates reaching 80.67% against Botrytis cinerea, 76.22% against Aspergillus niger and 66.67% against Fusarium culmorum for Serratia sp. The Halomonas sp. strain inhibited mycelial growth by producing volatile substances in 71.29% of niger, 75.49% of Aspergillus flavus and 72.22% of Penicillium glabrum. HPLC identification of the antifungal molecules by these three bacteria revealed that they , making these strains the first Rhizobacteria capable of phenolic compounds. Finally, pot tests were carried out to determine the effectiveness of these strains inpromoting wheat growth under salt stress (125 mM, 150 mM and 200 mM). The results showed that a consortium of two isolates (Serratia sp.-SB6 et Halomonas sp.-SB39) gave the best results at 125 mM with a significant increase in plant height of 30% on sterile soil and 46% on non-sterile soil. However, at higher concentrations, the halophilic sp bacteria gave the best results. In all cases, a significant improvement in the growth of wheat seedlings inoculated with the bacteria was observed compared to the non-inoculated . | |

L'application de bactéries et halophiles favorisant la croissance des plantes (PGPB : bactéries promotrices de la croissance des plantes) avec des activités de lutte biologique comme inoculant des plantes cultivées, offre une alternative biologique à l'utilisation des agrochimiques, particulièrement lorsqu’elles sont soumises à un stress salin. Dans cette perspective, 70 souches bactériennes ont été isolées à partir de sols salins (sebkha) dans des zones aride et semi-aride de l’est de l’Algérie. Trois isolats ont été sélectionnés sur la base de la production de molécules bioactives promotrices de la croissance des plantes. 100 % des isolats ont la capacité de fixer l’azote atmosphérique et de produire de l’ammoniac (NH3). 53,33% ont le pouvoir de solubiliser le phosphate inorganique, et 33,33% produisent du cyanure d’hydrogène (HCN) et 6,67% produisent l’AIA. Deux de ces isolats appartiennent au genre Serratia et le troisième est une bactérie halophile Halomonas. Ces bactéries ont été identifiées sur la base de la séquence de l’ADNr 16S. Des tests contre des champignons phytopathogènes ont été réalisés. L’identification les molécules antifongiques par ces bactéries a été faite par Chromatographie en phase liquide à haute performance. Ces bactéries sont capables d’inhiber le développement mycélien contre des champignons avec des taux atteignant 80.67 % contre Botrytis cinerea, 76.22 % contre niger et 66.67 % contre Fusarium culmorum pour Serratia sp. La souche Halomonas sp. a inhibé la par de substances volatiles de Aspergillus niger à 71.29 %, Aspergillus flavus à 75.49 % et Penicillium glabrum à un taux de 72.22 %. L’identification par HPLC des molécules antifongiques produites par ces trois bactéries révèle qu’il s’agit de , ce qui fait de ces souches les premières Rhizobactéries capables de produire des composés phénoliques. Enfin, des tests en pots pour déterminer l'efficacité de ces souches à promouvoir la croissance du blé sous stress salin (125 mM, 150 mM et 200 mM) ont été réalisés. Les résultats ont révélé qu’un consortium de deux isolats (Serratia sp.-SB6 et Halomonas sp.-SB39) avait donné le meilleur résultat à 125 mM avec une augmentation significative de la hauteur des plants de 30 % sur sol stérile et de 46 % sur sol non stérile. Mais, à des concentrations plus c’est la bactérie halophile Halomonas sp qui a donné le meilleur résultat. Dans tous les cas, on note une amélioration significative de la croissance des semis de blé inoculés avec les bactéries, comparés aux témoins non inoculés. | |
