Optimization of aeration rate and timing for ethanol production from date by-products by Saccharomyces cerevisiae using response surface methodology

dc.descriptionتعتبر التهوية هي واحدة من العوامل المهمة التي تؤثر على فسيولوجيا الخميرة وتؤثر على إنتاج الإيثانول .كان الهدف من باستخدام Saccharomyces cerevisiae هذه الدراسة هو تحسين معدل التهوية ووقت التهوية لإنتاج الإيثانول بواسطة 2.5 و ،1.5 ،0.5 ، تم تطبيق معدل التهوية ( 0.2 (RSM). نموذج مركزي مركب يعتمد على منهجية سطح الاستجابة 6 و 6.8 ساعة) مع معدل تحريك ثابت ( 120 دورة في الدقيقة) .كان وسط ،4 ،2 ، وتوقيت التهوية ( 1.2 ، (vvm2.9 إنتاج الإيثانول هو عصير التمر الذي يحتوي على 150 جرام/لتر من السكر الكلي و 1 جرام/لتر من اليوريا .تمت عملية التخمير عند درجة حرارة 30 درجة مئوية في مفاعل سعة 2 لتر .أظهر تحليل منهجية سطح الاستجابة أن التهوية المثلى و 1.2 ساعة، على التوالي .تحت الظروف المثلى، vvm لإنتاج الإيثانول كانت بمعدل تهوية ووقت تهوية قدرهما 1.5 82.74 جرام/لتر، 3.14 جرام/لتر/ساعة و 0.71 (Yp/s) وعائد الإيثانول (Qp) والإنتاجية ، (P) كانت تركيز الإيثانول هي 68.30 جرام/لتر، Yp/s و Qp و P جرام/جرام، على التوالي .في ظل ظروف عدم التهوية (معاملة التحكم)، كانت قيم 2.59 جرام/لتر/ساعة و 0.61 على التوالي .أظهرت النتائج بوضوح أن التهوية المثلى يمكن أن تحسن إنتاج الإيثانول
dc.description.abstractAeration is one of the significant factors affecting yeast physiology and impacts ethanol production. The aim of this study was to optimize the aeration rate and aeration time of ethanol production by Saccharomyces cerevisiae using a centered composite model based on response surface methodology (RSM). The aeration rate (0.2, 0.5, 1.5, 2.5 and 2.9 vvm), the aeration timing (1.2, 2, 4, 6 and 6.8 h) with a constant agitation rate (120 rpm) were applied. The ethanol production medium was date juice containing 150 g/L of total sugar and 1 g/L of urea. The fermentation was carried out at 30°C in a 2-L fermenter. Response surface methodology analysis showed that the optimal aeration for ethanol production was an aeration rate and aeration time of 1.5 vvm and 1.2 h, respectively. Under the optimal conditions, the ethanol concentration (P), productivity (Qp) and ethanol yield (Yp/s) were 82.74 g/L, 3.14 g/L/h and 0.71 g/g, respectively. Under an unaerated condition (control treatment), the P, Qp and Yp/s values were 68.30 g/L, 2.59 g/L/h and 0.61 respectively. The results clearly indicated that optimal aeration can improve ethanol production.

L'objectif de cette étude consiste à optimiser le débit et le temps d'aération de la production d'éthanol par Saccharomyces cerevisiae à l'aide d'un modèle composite centré basé sur la méthodologie des surfaces de réponse. Le débit d'aération (0.2, 0.5, 1.5, 2.5 and 2.9 vvm), le temps d'aération (1.2, 2, 4, 6 and 6.8 h) avec une vitesse d'agitation constante de 120 rpm ont été appliqués. Le milieu de production d'éthanol à base de jus de datte contenant 150 g/L de sucres totaux et 1 g/L d'urée. La fermentation a été réalisée à 30°C dans un fermenteur de 2 L. L'analyse de la méthodologie des surfaces de réponse a montré que les conditions optimales pour améliorer la production d'éthanol étaient avec un débit d’aération de 1,5 vvm et un temps d'aération de 1,2 h. Dans ces conditions, la concentration en éthanol (P), la productivité (Qp) et le rendement en éthanol (Yp/s) étaient respectivement de 82,74 g/L, 3,14 g/L/h et 0,71 g/g. Dans des conditions non aérées (témoin), les valeurs de P, Qp et Yp/s étaient respectivement de 68,30 g/L, 2,59 g/L/h et 0,61 g/g. Ces résultats indiquent clairement qu'une aération optimale peut améliorer la production d'éthanol.
