Abstract
Capparis spinosa is a Mediterranean shrub belonging to the Capparaceae family, widely used in traditional herbal medicine. This plant has several biological activities, such as antioxidant, hepatoprotective, and immunomodulatory effects. The objective of this study is to evaluate the antioxidant and anti-inflammatory effects of methanolic extracts of C.spinosa from leaves (EMF), stems (EMT), and fruits (EMFr).The determination of total polyphenols, flavonoids, and tannins shows that EMF contains the highest concentrations of polyphenols (57.37 ± 0.9 µg AG Eq/mg of extract), flavonoids (31.34 ± 9.15 µg Q Eq/mg of extract), and tannins (275.11 ± 2.51 µg AT Eq/mg of extract). This extract also showed the highest reducing power (EC50 = 32.83 ± 0.57 µg/ml) and the highest scavenging activity against DPPH and ABTS radicals (IC50 = 30.51 ± 1.86 µg/ml and 11.93 ± 0.08 µg/ml, respectively). The prevention of β-carotene bleaching was almost similar for the three extracts studied. The evaluation of the anti-edema effect shows that EMF inhibited xylene-induced edema in mice by 84.86% at a dose of 600 mg/kg. EMFr was effective against croton oil-induced edema in mice (35.89%) and carrageenan-induced edema in rats (52.50 ± 17.27% at 3 hours) at a dose of 300 mg/kg. Finally, an anti-inflammatory effect was also obtained preventively with EMFr and EMF (41.71% and 39.74% respectively) in the Freund's complete adjuvant-induced arthritis (AIA) model in rats. This effect resulted in an improvement in clinical, serum, and histological parameters. These results confirmed that parts of C. spinosa have various antioxidant and anti-inflammatory effects linked to their polyphenol and/or flavonoid content and justify the widespread traditional use of this plant. C. spinosa extracts may therefore be a good source of phytoconstituents used as drugs to treat inflammatory diseases.
