Abstract
This PhD thesis focuses on the design, synthesis, and evaluation of new α-aminophosphonic acids (HAP1 and HAP2) as promising bioactive compounds. It begins with a comprehensive review of organophosphonates, emphasizing their importance in medicinal chemistry due to the stability of the carbon–phosphorus (C–P) bond, which allows them to mimic biological phosphates and act as enzyme inhibitors. The experimental work involves the synthesis of the target compounds, followed by their characterization using techniques such as FT-IR, UV-Vis, and NMR spectroscopy. In addition, computational studies based on Density Functional Theory (DFT) and molecular docking were conducted to understand their structural, electronic, and biological interactions, alongside ADME-T predictions to evaluate their pharmacokinetic and toxicity profiles. The biological investigations demonstrated that the synthesized compounds exhibit significant antioxidant activity, supported by multiple assays (DPPH, ABTS, FRAP), as well as synergistic effects when combined with standard antioxidants. Furthermore, they showed antibacterial properties and enzyme inhibition potential. A key innovation of this work lies in the incorporation of these compounds into hyaluronic acid-based nanohydrogels, enabling controlled drug release, improved stability, and enhanced bioavailability. Overall, this study highlights the potential of α-aminophosphonic acids as valuable candidates in pharmaceutical development, particularly when integrated with nanotechnology-based delivery systems.

Cette thèse de doctorat porte sur la synthèse et l’évaluation de nouveaux acides α-aminophosphoniques (HAP1 et HAP2) en tant que composés bioactifs prometteurs. Elle débute par une revue bibliographique approfondie des organophosphonates, mettant en évidence leur importance en chimie médicinale grâce à la grande stabilité de la liaison carbone–phosphore (C–P), qui leur permet d’imiter les phosphates biologiques et d’agir comme inhibiteurs enzymatiques. Le travail expérimental comprend la synthèse des composés cibles, suivie de leur caractérisation à l’aide de techniques spectroscopiques telles que FT-IR, UV-Vis et RMN. Par ailleurs, des études computationnelles basées sur la théorie de la fonctionnelle de la densité (DFT) et le docking moléculaire ont été réalisées afin de mieux comprendre leurs propriétés électroniques et leurs interactions biologiques, complétées par des prédictions ADME-T pour évaluer leurs profils pharmacocinétiques et toxicologiques. Les résultats biologiques ont montré que les composés synthétisés présentent une activité antioxydante significative, confirmée par plusieurs tests (DPPH, ABTS, FRAP), ainsi que des effets synergiques en combinaison avec des antioxydants standards. Ils ont également démontré une activité antibactérienne et un potentiel d’inhibition enzymatique. Une contribution majeure de cette étude réside dans l’incorporation de ces composés dans des nanohydrogels à base d’acide hyaluronique, permettant une libération contrôlée, une meilleure stabilité et une biodisponibilité accrue. Dans l’ensemble, ce travail met en évidence le potentiel des acides α-aminophosphoniques comme candidats prometteurs en développement pharmaceutique, notamment grâce à l’intégration des nanotechnologies.
