Abstract

The aim of this study is to determine the chemical composition, phenolic content, as well as the antibacterial, antifungal and antioxidant activities of hydromethanolic extracts from R. chalepensis collected at three different periods: before flowering (BF), during flowering (DF) and after flowering (AF). Three hydromethanolic extracts were prepared: the before flowering extract (BFE), during flowering extract (DFE) and after flowering extract (AFE). Hydromethanolic extracts analysis was conducted regarding their chemical composition by HPLC-DAD-ESI/MS method, quercetin, graveolin, bergapten, isoimperatorin, chalepin, rutamarin and β-carotene were detected. Also, total phenolics and flavonoids contents were determined. The AFE displayed the maximum polyphenol and flavonoid levels; 84.54 ± 1.66 µg GAE/mg dry extract and 25.15 ± 0.88 µg QE/mg dry extract, respectively. Antibacterial activity was evaluated against seven bacterial strains using disc diffusion (DD) method and minimal inhibitory concentration (MIC). The results of the studied plant showed an important antibacterial activity against both Gram-negative and Gram-positive bacteria. The bacterial strains showed sensitivity ranging from moderate to high, either in terms of ID (08.67 to 17.67 mm) or in terms of MIC (01.95 and 31.25 mg/ml). These extracts also exhibited excellent activity against eight filamentous fungi growth when tested by incorporation of plant extracts into the culture medium method at (10%) reduced the growth from 25 % to 97% of all fungi. On the other hand, The antioxidant activity was assessed using four methods; DPPH ,with IC50 ranging between 14.18 ± 1.02 and 30.96 ± 1.80 μg/mL, β-carotene/linoleic acid, with IC50 ranging between 34,86 ± 0,78 and 49,06 ± 0,15 μg/ml and reducing power, with EC50 varied between 100,61± 1,25 and 139,99± 2,08 µg/ml and TAC, with values ranging between 125,12 ± 2,47 and 160.59 ± 5.78 µg AAE/mg extract. In conclusion, the results revealed that R. chalepensis hydromethanolic extracts have strong antibacterial and antioxidant properties, highlighting their potential as natural sources for developing novel antibacterial and antioxidant agents

L’objectif de ce travail est l’étude de la composition chimique, la teneur en composés phénoliques ainsi que les activités antimicrobiennes et antioxydantes d'extraits hydrométhanoliques de Ruta.chalepensis (R. chalepensis) récolté pendant trois périodes différentes : avant (AVF), pendant (PF) et après (APF) floraison. Trois extraits hydrométhanoliques sont alors préparés. L’analyse chimique par HPLC-DAD-ESI/MS a permis d’identifier les métabolites secondaires suivants : quercétine, graveoline, bergapten, isoimperatorin, chalepin, rutamarin et β-carotene dans les trois extraits. L’extrait EAVF a présenté des teneurs en polyphénols et en flavonoïdes de 84,54 ± 1,66 µg EAG/mg d'extrait sec et 25,15 ± 0,88 µg EQ/mg d'extrait sec respectivement. L'activité antibactérienne a été réalisée sur sept souches bactériennes par la méthode de diffusion sur disque (DD), la concentration minimale inhibitrice (CMI) et la concentration minimale bactéricide (CMB) ont été déterminées. Les résultats ont montré une sensibilité bactérienne importante des bactéries Gram- et Gram+ qui ont montré des Ǿ (08,67 à 17,67 mm) soit en termes de CMI (01,95 et 31,25 mg/ml) et des CMB (3,9 et 62,5 mg/ml). L’activité antifongique a été évaluée sur 8 souches de champignons de contamination alimentaire et phytopathogènes, deux méthodes ont été appliquées sur milieu solide, les extraits de R. chalepensis ont montré un effet d’inhibition important vis-à-vis des souches testées allant de 25 % à 97% d’inhibition. L’activité antioxydante a été évaluée selon quatre méthodes. A l’issue des tests d’activité antioxydante in vitro, les EAVF, EPF et EAPF ont montré un effet scavenger très puissant du radical DPPH avec des valeurs de IC50 qui varient de 14,18 ± 1,02 à 30,96 ± 1,80 µg/ml ; Ces extraits ont également inhibé l’oxydation du β-carotène, notamment l’EAPF avec un pourcentage d’inhibition de 49,06 ± 0,15, concernant la capacité antioxydante totale CAT; L’EPF a exercé la plus forte capacité antioxydante (160,59 ± 5,78 µg AAE/mg d'extrait sec). Aussi, les extraits ont exhibé de puissants pouvoirs réducteurs du fer avec des valeurs de 100,61± 1,25 à 139,99± 2,08 µg/ml. En conclusion, les résultats ont révélé que les extraits de R. chalepensis possèdent de fortes propriétés antibactériennes, antifongiques et antioxydantes, soulignant leur potentiel comme sources naturelles pour le développement de nouveaux agents antibactériens et antioxydants
